Q: do I have to purchase on a regular basis or can this be a one off purchase

You do not need to set up a subscription. I purchase a 6 pack and it lasts me for 2-3 years. I use it daily on my full upper's and have not yet tried it on my partial lowers. It's a super product and is better than the 'glue'. No oozing....yay! It takes a few times using it to see how much you like to use.

Q: Is there zinc in this ?

The is no Zinc in it, that is why I use it.

Q: Is this suitable for a partial lower front denture with one tooth on?

I use the Poligrip powder on a full upper denture but on my lower partial I use the Poligrip Denture Adhesive Cream which works very well for me.
