It is based in total coolant volume. If your vehicle's coolant capacity is higher than the recommended maximum dilution quantity, you will need to purchase another.

Is this a 10 minute flush or does it stay in longer?

Follow the instructions on the bottle. It is not recommended to drive with it in nor have multiple engine on/off cycles. Letting the car idle at least 10-15 minutes will not hurt the system, but do not be excessive.
