What size of tube will fit through the hole?

The diameter of the hole in MYLOLF water jug cap is 18mm, so tubes with a diameter lower than 18mm can be passed through.

Will this work with a 53mm opening?

You will have to make the decision....a 3 or 5 gallon water jug has about a 48mm opening. The silicone is flexible...if you purchase it and can return it if it does not fit,,,,then go for it.

How long do they last before you have to replace them?

I don’t know that answer. They have held up pretty well so far. They are also super easy to use and stay on very well.
