Q: I need to drill to 3/8" in-line holes (along the length) through 1" steel tube? Is this the tool I start with?

Yes. Use a pipe vise to secure the pipe. Level the centering tool and Mark by hitting at both ends of the pipe. Snap a chalk line the mark centers for all of your holes.

My order only had 1. My order was more than two years ago.

Q: Pipe is generally a term used for id description, will this work on 1/2" od stainless tubing?

1/2 " OD is the very minimum it will work on in my experience. 1/2" will not fit on the wings, but will center on the inside of the edge of the wings. It does not work as well as on larger diameter work, but will find center.
