What does speaker level input & signal sensing turn on do? I see the plug on the front of the amp.

This is the Speaker Level Input adapter. This is only for people who will connect their amps to a factory system by splicing into your rear speaker wires.If you are using RCA cables to feed signal to your amplifier, you don't need it. Keep it in case you need to return or RMA the amplifier though.

Would this be good to power one 12" subwoofer? My old amp was a Crossfire 600W.

If the subwoofer is rated at 300 watts or less I would say yes. Generally speaking the wattage of the amp should be greater than the wattage of the speaker to sufficiently power it.
