Doed this have a designated spot to padlock the box shut?

It does NOT. I removed the clasp and bought a lockable one at Home Depot. Small locks are available from Amazon. Then touched up the small amount of the exposed wood with a furniture marker pen. Worked out fine.

Would this hold a regular size envelope (letter sized)?

Yes it will. Inside dimensions are 10 x 6 1/4. Envelope is 9 1/2 x 4 1/8

Can a wine bottle fit inside?

Not a regular-sized one, maybe one that is fairly stubby.
