Q: can bowl be filled and then flushed

No, but the bowl does collect about 1-2 inches of water in the bottom. When the bottom seals up the water still runs for a second or two and the water stays in the bottom for upto a week with out draining out. You can also let more water in by slighly touching the foot pedal allowing water to come in to the bowl but not opening the seal. So it will not work like your house toliet but it does have a great seal that will allow you to manipulate it to funtion almost like your house toliet.

Q: Can the seat be replaced with a standard toilet seat?

It does have standard width mounting holes for the toilet seat so you could replace the seat with anything you want from a home improvement store.

Q: Does this unit come with a closet flange seal or does that have to be purchased separately?

The toilet comes with a new seal and flange bolts.
